All organelles in a higher organism - the nucleus, mitochondria, chloroplasts, endoplasmic reticulum (ER), and others - perform specific functions and must therefore contain a certain set of proteins. Most of these proteins are synthesized in the cytosol, but specific "zip codes" or "signal sequences" direct them to their different final destinations. During their transport, many of these proteins have to traverse at least one membrane or need to be integrated into a membrane. The mechanisms by which signal sequences are recognized and by which proteins are transported is best understood for the endoplasmic reticulum (ER). Structural and biochemical data show that signal sequences open a protein-conducting channel, allowing polypeptides to move across or integrate into the membrane. Organelles also have characteristic shapes that are generated by specific proteins, and the mechanisms by which they function are being unraveled.
