The Jak (Janus kinase)-Stat (signal transducer and activator of transcription) signalling pathway plays a central role in normal organ development as well as in the development of leukemias and carcinomas. Lung cancer, breast cancer and brain tumors e. g. are triggered by excessive activation of Stat3 and Stat5. Activated Stat molecules form dimers and assume the function of sequence specific transcription factors in the cell nucleus. The induction and suppression of target genes leads to celltype specific effects. In cancer cells, activation of Stat3 and Stat5 results in enhanced cell proliferation, resistance to apoptotic signals and secretion of angiogenic or immunosuppressive factors. A control function of Stats in lysosome-mediated cell death and tumor cell metabolism has only recently been discovered. Due to the many signals that can cause Stat activation, e. g. growth factor receptors, cytokine receptors, but also cytoplasmatic tyrosine kinases, inhibition of Stats in tumor cells has proved difficult so far. Although siRNA interference studies have shown Stats as promising targets, specific drugs are still in very early stages of development.
