Abstract: Intracellular protein adaptor domains play an ubiquitous role in the formation of proteins assemblies inside the cell. With a focus on proline-rich sequence recognition domains phage display was used to decipher the recognition code of several of these domains. Subsequently it could be shown that peptides derived from these studies represent useful tools in SILAC/MS experiments that are meant to decode the corresponding interactomes. Furthermore, certain adaptor domains can form the basis to design scaffolds with novel binding properties. Finally, sortase-mediated protein ligation can be invoked to investigate multidomain adaptor proteins in regard to specific structural and mechanistic questions.
