Abstract: DNA of higher organisms is highly compacted within the cell nucleus. DNA is wrapped around units of specialized proteins, forming cylindrical structures termed nucleosomes. This bead-on-a-string like structure called chromatin condenses further into chromosomes. While the structure of the DNA itself is known on an atomistic level, the higher level folding of chromatin remains enigmatic despite extensive research. We tackle this problem by using Monte Carlo-simulations applying Replica Exchange and Simulated Annealing procedures to integrate experimental knowledge from heterogeneous sources like single molecule experiments and high throughput sequencing. In this talk I will present our models, computational approaches and its applications in biophysics and systems biology.
