Neue Strategien für die Expression funktionaler Membranproteine in E.coli

Das Institut für Biochemie lädt zu einem Kolloquium am 18. November 2013 ein. Dr. Jan Kubicek, Cube Biotech GmbH, Monheim, referiert zum Thema: "Neue Strategien für die Expression funktionaler Membranproteine in E.coli".

Abstract: Membranproteine machen ca. 30% des humanen Genoms aus, und stellen über 50% der pharmazeutisch relevanten Zielproteine dar. Trotzdem sind bisher nur wenige Daten zur Struktur und Funktion von Membranproteinen verfügbar, da die Expression und Reinigung dieser Proteinklasse schwierig ist und nur wenige etablierte Protokolle verfügbar sind. Wissenschaftler bei Cube Biotech haben diese Protokolle über Jahre etabliert und optimiert, so dass wir in der Lage sind, funktionale humane Membranproteine zu exprimieren, zu reinigen und funktional zu rekonstituieren, z.B. in Nanodiscs. Als Wirtsorganismus verwenden wir meist bakterielle E.coli Stämme, da sie innerhalb kurzer Zeit bei moderaten Kosten Protein produzieren. Wir werden Daten zur funktionellen Charakterisierung von humanen GPCRs und anderen Membranproteinen präsentieren, und auch erste Erfolge der Strukturaufklärung durch Kristallisation in einer von uns entwickelten Variante der lipidisch-kubischen Phase (CIMP) zeigen.

Approximately 30% of a genome encodes for membrane proteins, one of the therapeutically most important classes of proteins that represents more than 50% of known and novel drug targets. Progress in this area is slowed by the lack of methods and procedures for expression and isolation of membrane proteins in quantities which allow their functional characterization and structural analysis. We have developed a complete workflow of optimized protocols for expression, isolation and reconstitution of functional human membrane proteins e.g. into nanodiscs. We are using E.coli as an expression host whenever possible to profit from the high protein yields these bacteria can produce in a short time at moderate cost. We will present functional characterization data for human GPCRs and other membrane proteins as well as initial data towards their structure determination by crystallization in a modified version of lipidic cubic phases (CIMP) that was developed together with our researchers.
