Abstract
During the last decade the effect of neoplastic transformation on the metabolism and the knowledge about the molecular mechanisms of the cellular adaption to hypoxia has increased, but there exists little information about the overlap and differences in gene/protein expression of both processes as well as the consequences on the tumor microenvironment, immune modulatory molecules and immune responses. Therefore the aim of this study was to dissect oncogene- and tumor suppressor-mediated and hypoxia-regulated alterations on (i) the metabolism of tumor cells, (ii) the tumor microenvironment and (iii) immune responses. In addition, the observed alterations were modulated/reversed by drugs and small molecules. Using different experimental approaches including ome-based strategies the effect of oncogene-/tumor suppressor gene- and hypoxia-regulated altered gene/protein expression pattern on the cellular metabolism, immune responses and their modulation by diverse substances was analysed by determination of glucose uptake, lactate secretion, extracellular pH, lactate dehydrogenase activity, amino acid content, ATP levels and T cell activity.
