Abstract: Redox modifications have long been discussed to be the result of increased levels of pro-oxidants, for instance due to irradiation or decreased levels of antioxidants, conditions best known under the term “oxidative stress”. Today, the field is experiencing an exciting transformation, from looking at a general redox homeostasis and the pathological oxidative stress model to realizing redox changes as part of localized, rapid, specific and reversible redox-regulated signaling events, despite of the highly transient and volatile nature of redox modifications that makes them very difficult to access for broad in-depth investigations. Proteins can be regulated post-translationally via reversible redox modifications of susceptible amino acid side chains or cofactors. The thiol groups of cysteinyl side chains constitute the major targets. In response to specific oxidants, susceptible thiols can be oxidized to form protein disulfides, others undergo S-glutathionylation, S-nitrosylation, or sulfenylation. These modifications are enzymatically regulated by proteins of the Thioredoxin (Trx) family, i.e. Trxs, glutaredoxins (Grxs), and peroxiredoxins (Prxs). Consequently, these proteins are increasingly recognized for their specific role in redox signaling. Our main interest is the identification and characterization of such signaling events in various physiological and pathological settings. In this presentation, we will present the new concept of redox signaling, and provide an overview of our recent results demonstrating its importance in the fields of neuronal development, cell migration and cancer progression, iron homeostasis, apoptosis, and ischemia-reperfusion injury.
