Abstract: Synaptic transmission is triggered by SNARE protein mediated membrane fusion. A coiled-coil four-helix bundle is formed between proteins residing in the plasma membrane and in the membrane of synaptic vesicles, forcing the two merging membranes in close proximity. Artificial SNARE mimicking model systems are explored with respect to the fusion mechanism. The participation of transmembrane domains in the fusion process, the control of docking vs. fusion, and various recognition motifs with the potential to imitate a zipper-like recognition are investigated applying SNARE analogous model systems. All analogs contain transmembrane peptides and covalently linked recognition motifs. Coiled-coil forming peptides[1] and various kinds of peptide nucleic acids[2] are described for recognition allowing systematic structural variations and an overall simplified SNARE assembly mechanism. Artificial SNARE mimicking model systems are reconstituted in vesicles and docking or fusion efficiency is reported.
