Abstract:
As potential candidates for designing new drugs against tuberculosis, the genes encoding LeuD, LeuC, LeuB and IlvE, involved in leucine biosynthesis in M. tuberculosis were targeted for structure determination.
All four targeted genes were sucessfully cloned, expressed in soluble form in E. coli expression hosts. The structure of LeuB was determined in apo-form, in complex with substrate (Isopropylmalate) and in complex with Mg-ion. High throughput screening resulted in identification of an inhibitor with IC50 of about 35 micromolar.
