The correct functioning of host innate immune responses to viral infections is based on reliable pathogen detection and is essential in limiting pathogen replication and spread. The presence of viral RNA activates cellular RNA sensors and results in expression of cytokines and type I interferon (IFN-I). IFN-I in turn signals to express an array of genes that restrict viral growth. Using coronavirus models of infection we study the importance, dynamics and specificities of IFN-I responses to cytopathic RNA virus infection. We focus in particular on important target cells of virus infections, such as dendritic cells and macrophages, and on the major producers of IFN-I during the early phase of the infection, namely plasmacytoid dendritic cells.
