ω-Transaminases (EC 2.6.1.18) are attractive biocatalysts for synthesis of chiral amines. These enzymes catalyze transfer of an amino group from a donor substrate to an acceptor carbonyl compound using the cofactor pyridoxal-5´-phoshate (PLP). ω-Transaminases do not require a carboxylic acid group in a-position (in contradiction to a-transaminases) and hence accept a wider spectrum of ketones or amines.
Our homology model-based rational design approach will be discussed in the lecture including some results to convert an S-selective ω-transaminase variant from Arthrobacter citreus to an R-selective variant. This homology modelling/rational design approach has further been applied to optimize ω-transaminase from Chromobacterium violaceum.
