Optical Sectioning and beyond: Possibilities and Advances

The workshop “Optical Sectioning and Beyond: Possibilities and Advances in Light Microscopy” will examine issues important to both the cell and neurobiologist but also to users of modern lightmicroscopical procedures in general. Drawing on current R&D projects, the workshop will illustrate the advantages and potential of modern lightmicroscopy and describe new trends in development.

Today, highly-developed light-microscopical techniques are indispensable in conducting biomedical and natural science research. Being “enabling technologies”, they allow insight into biological processes which, till now, had only been possible through conventional cell biological and biochemical methods.
With the help of confocal microsocopy and immunocyto- and histological techniques involving fluorescent biomolecules and “tags”, highly-complex cellular relationships and structures can now be displayed as active, 3-dimensional reconstructions.
Speed and temporal resolution of signal detection as well as computer-based signal processing have greatly increased over the years, with the focus being on the conservation of living samples. Thus, even 4-dimensional analyses have become possible.
Moreover, applications such as FLIM, FRET and FRAP have reached incredible fidelity. The dynamics of biological processes in time and space are taken into account in all these developments.
Methods of superresolution in light microscopy have begun to blur the boundaries with electron microscopy. Light microscopical
imaging follows the development of biological detector-probes whose physical dimensions are in the lower nanometer scale. The detection of single biomolecules within their biological
surroundings has become a reality.
Through in vivo microscopy methods, cellular processes can be observed under natural circumstances. The key elements of in situ imaging are high penetration depth, tissue protection and miniaturisation of optical components. These technologies hold exciting possibilites for the future: pathological alterations can be detected over time or cellular activity involved in highly complex
behavioural patterns can be identified.
