Abstract: „We have recently designed lipase congeners by global substitutions of canonical amino acids (cAAs) with corresponding non-canonical counterparts (ncAAs) in residue-specific manner. Expectedly, we have found that different ncAAs carrying various chemical modifications (e.g. fluorination, methylation, hydroxylation, etc.) dramatically altered the activity of lipase from the thermophilic anaerobic bacterium Thermoanaerobacter thermohydrosulfuricus (TTL). Observed effects include significantly enhanced activity, markedly extended substrate tolerance, large changes in optimal temperature and pH values."
