Der Inhibitor Ex-527 bindet einerseits an das Enzym Sirt3 (hellgraue Oberfläche), andererseits an acetylierte ADP-Ribose; diese Substanz ist ein Produkt der von Sirt3 zunächst ausgelösten Deacetylierung. In der abgebildeten Konstellation ist das Aktivzentrum des Sirtuins blockiert; das Sirtuin bleibt daran gehindert, weitere Deacetylierungsprozesse in Gang zu setzen.
