Bei Nagern führt eine periphere Nervenverletzung oder Entzündungsreaktion zu einer Hochregulation der GTP Cyclohydrolase (GCH1) und Steigerung der Tetrahydrobiopterin (BH4) Produktion. BH4 führt durch Steigerung des neuronalen Calciumeinstroms und der Stickstoffmonoxid (NO) Freisetzung zur Schmerzauslösung. Wird die Enzymaktivität der GCH1 gehemmt, werden BH4 Produktion, NO-Freisetzung und Calciumeinstrom normalisiert und die Schmerzen deutlich reduziert. Bei Menschen gibt es eine genetische Variante der GCH1, die eine exzessive BH4 Produktion verhindert und dadurch chronische neuropathische und akute Schmerzen reduziert. Der Zusammenhang mit Tumorschmerzen wird in diesem Projekt untersucht.
