p53 ist ein Schlüsselprotein mit enormer Bedeutung. Bei Schädigung der DNS – etwa durch Bestrahlung oder spontane Mutationen – wird es vermehrt exprimiert. In der Folge wird der Zellzyklus angehalten und der programmierter Zelltod ausgelöst, um Schlimmeres zu verhindern. Fehlt dieser Tumorsuppressor, beginnen sich Zellen mit einer geschädigten DNA unkontrolliert zu teilen; es entstehen Tumore.
Vousden und ihr Team versuchen mit Hilfe von bestimmten Medikamenten, die in Krebszellen verloren gegangene p53-Funktion wiederherzustellen. Die Forscher hoffen, das Krebswachstum auf diese Weise aufhalten und die entarteten Zellen töten zu können.
