Außen dran statt innen drin: Die Graphik zeigt zwei Chromosomen (blau) in schematischer Darstellung. In den Chromosomen ist die Erbsubstanz DNA in hoher Dichte auf Protein-Körperchen aufgespult. Fremde DNA-Elemente (rot) können sich auf verschiedene Weisen mit einem Chromosom verbinden: Ein gängiger Weg ist die Integration in das Chromosom, wobei sie zu einem festen Bestandteil, einem Abschnitt von ihm, werden (untere Bildhälfte). So schleusen sich viele Viren in die DNA ihrer Wirtszelle ein, und so arbeiteten auch die klassischen Vektoren (DNA-Transportelemente) für die Gentherapie. Anders Episomen (obere Bildhälfte): Diese ringförmigen DNA-Moleküle heften sich nur von außen an bestimmte Träger-Strukturen des Chromosoms an. Der Vorteil: Dabei werden die Gene innerhalb des Wirts-Chromosoms nicht geschädigt. Deshalb versuchen Forscher jetzt, Episomen für die Gentherapie nutzbar zu machen.
