Natural killer (NK) cells represent a discrete cell population of the innate immune system. Although they share a common killing mechanism with cytotoxic T lym-phocytes of the adaptive immune response, their function and the recognition principles of target cells (e. g. cancer cells and virus infected cells) are entirely different. NK cells display a broad variety of germ-line encoded activating and inhibitory re-ceptors whose downstream signal is inte-grated as the basis to decide whether the target cell should be regarded as "normal self" or "abnormal/non-self". In the latter case the target cell will be destroyed. Although this scheme looks rather trivial, important cellular ligands on the target cell have not been identified yet nor have NK cell education and the integration process of receptor signalling been understood. Most recently, NK cells were shown to generate an immunological memory, thus opening up entirely new perspectives. Accumulating evidence shows that NK cells can eliminate tumours and control herpes-virus infections in an early state, contribute to the development of autoimmune diseases and regulate the immune response mediated by dendritic cells, macrophages and T cells. Therefore, NK cells are an appealing target and effector in human therapy.
