Abstract:
Biofilm systems are microbial communities associated with interfaces. They comprise bioaggregates (mobile) and biofilms (stationary). Biofilm systems are of major importance in three different areas:
1. Biofilms in the environment
2. Biofilms in technical systems
a) wanted - biotechnology, wastewater treatment
b) unwanted - biofouling, biodegradation
3. medically relevant biofilms
Major progress in understanding microbial biofilm structure and function has been made by using laser scanning microscopy. The advantages of laser scanning microscopy include the analysis of 3-dimensional fully hydrated samples and multi-channel imaging in combination with digital image analysis. By means of laser scanning microscopy the samples can be analysed for their reflection and autofluorescence properties. The availability of many different fluorescence probes allow the investigation of biofilm development and detachment, biofilm structure and composition and biofilm processes. In addition, the biofilms can be examined with respect to the distribution of bacterial cells and extracellular polymeric substances, the identity of bacterial groups or species, the activity of bacteria and enzymes as well as the biofilm microhabitat. The 3-dimensional digital image data sets recorded by laser scanning microscopy can finally be visualised and quantified and may serve as a basis for biofilm modelling.
In the first part of the presentation a short introduction into biofilm systems will be given. The second part will cover the applicability of advanced laser scanning microscopy and digital image analysis for studying interfacial microbial communities in various habitats.
